How can you position two people under a 12x12 tarp?
Any way you want, that is a lot of room for two people. I use a 10X12, and my son and I fit under it very nicely. Most of the time we use 2 trees, side by side, but it works well with 3 trees too. I use my poles to make a canopy on one side, and this allows more room when using 3 trees.

My wife and I like the bunk-bed approach. We set one low and one quite high. With a small tarp you wouldn't have enough side coverage for the person on bottom with this setup but a 12x12 would be fine.
As long as you're talking about a 12x12 square and not diagonal measurements, you should have plenty of room to do whatever. I like to hang from either three trees or two if one tree is large enough in diameter to hang the hammock straps on both sides of the tree. Using two trees keeps the ridglines of the tarp centered better, with three trees there will be more coverage over one hammock than the other, unless you tie the tarp to another line that is run between the trees.
There are ways to use a spreader bar (i.e. Stick) to spread the hammocks apart if you have only two trees and they aren't large enough to spread the hammocks, but one hammock will move when the other is loaded/unloaded. This isn't a big issue if you use a structural ridgeline, because the hammock's sag won't change.
